When you finish making your changes, export the rosters to your data database folder (not the data folder). When you start up the game, these will be the default rosters that are pulled in.

MVPEdit does not allow you to create .SAV file which are pulled into the game from the "Load Rosters" screen.

You cannot load a .mbe file into the game. The .mbe files are only used by MVPEdit.

With this in mind, when starting a dynasty, should the "Use default rosters" prompt be answered yes or no? Something I've always wondered.....
